The Nikon A1 is the older Nikon confocal laser scanning microscope at the AMF, sharing the Nikon Ti2 body with the Abbelight SAFe-360 super-resolution platform via the LU4A beam switcher. Users can run confocal acquisitions on the A1 or switch to SMLM on the Abbelight on the same sample, individually or in sequence, on a single Ti2 stand. The A1 runs Nikon NIS-Elements AR.
Configuration
- Stand: Nikon Ti2 inverted microscope (shared body with Abbelight SAFe-360)
- Scan head: Nikon A1 (galvo and resonant scanning)
- Beam switcher: LU4A pad, switches between confocal and Abbelight light paths
- Excitation channels: 405, 488, 561, 640 nm (4 channels via the LU4 laser combiner; up to four fluorophores)
- Detectors: DU4 spectral detector unit (4-channel) and SD/VF detectors
- Z-control: Step-by-step Nikon A1 piezo Z, Perfect Focus System (PFS)
- Objectives: 60× TIRF, 100× TIRF (objectives are shared with the Abbelight platform on the same body)
- Live-cell: Okolab Top-stage Incubator (temperature, humidity, CO2)
- Software: NIS-Elements AR 5.21.03

Status
Operational. Older Nikon confocal generation, retained as the everyday confocal at the AMF alongside the Abbelight super-resolution platform on the same body. The newer Nikon A1R HD25 is hosted separately by the Soft Matter and Biological Physics group in Flux.

Trained on this instrument?
Read the Nikon A1 NIS-Elements MVP before your first solo session, then book via Calira. New users: start with the training pathway.
Contact
Day-to-day operation and user training are coordinated by the AMF team. For questions about the A1, contact Bianca van Driel (Lab Manager) at b.f.v.driel@tue.nl, or write to superresolution@tue.nl.
