An interferometric scattering (iSCAT) microscope for label-free imaging of single proteins, vesicles, viruses, and nanoparticles. The setup supports mass photometry, which determines the mass of individual proteins in solution from how strongly they scatter light, and iNTA (interferometric Nanoparticle Tracking Analysis), which measures size and refractive index of nanoparticles in suspension down to a few tens of nanometres. Both work without fluorescent labels.
The system has a 638 nm iSCAT channel and 488 nm and 561 nm excitation for fluorescence (coming soon). Because we are building it ourselves, we can swap parts, add channels, or change the layout as needed.
Capabilities
- Mass photometry of single proteins in solution
- iNTA sizing of nanoparticles, vesicles, and viruses
- Label-free single-particle imaging at 638 nm (488 and 561 nm in development)
- Two-colour fluorescence at 488 and 561 nm (in development)
- Correlative iSCAT and fluorescence imaging (in development)
Located at
Single Particle Metrology, MBx, FLUX building, TU/e campus. Visitors welcome on request.
What is coming
Mass photometry and iNTA are being completed first. After that we will bring in the 488 nm and 561 nm fluorescence channels and combine them with the iSCAT channel for correlative imaging. Further additions will depend on what experiments come up.
